Real Time Polymerase Chain Reaction (PCR) Facility

Principle
This facility enables researchers to perform highly sensitive, accurate, and reproducible quantitation of gene expression. The qRT-PCR systems can be used with TaqMan probe-based chemistry or intercalating dyes such as SYBR Green. The qRT-PCR monitors the amplification of a targeted DNA molecule during the PCR i.e. in real-time, and not at its end, as in conventional PCR. Real-time PCR is carried out in a thermal cycler with the capacity to illuminate each sample with a beam of light of at least one specified wavelength and detect the emitted fluorescence by the excited fluorophore.
The TaqManTM probes and SYBER Green are used for detection of PCR products in real-time PCR machine. SYBER Green is non-specific fluorescent dyes that intercalate between double-stranded DNA and TaqManTM probes are sequence-specific DNA probes labelled with a fluorescent reporter, taqman probes permits detection only after hybridisation of the probe with its complemetry sequence.

Specifications
The real-time machines are compatible with the following standard dye sets: FAM/SYBR Green I, VIC/JOE, NED/ TAMRA/ Cy3, ROX/Texas Red, and Cy5 dyes. SYBR green assays. Multiplexing: Five dyes per well. Mx3005P, CFX96 & 7500 Fast are 96 wells plate qPCR machine while in QuantStudio 6  user can use both 96 wells and 384 wells plate.
